Itchiness explained: Specific set of nerve cells signal itch but not pain, researchers find

Jan. 2, 2013 ? Johns Hopkins researchers have uncovered strong evidence that mice have a specific set of nerve cells that signal itch but not pain, a finding that may settle a decades-long debate about these sensations, and, if confirmed in humans, help in developing treatments for chronic itch, including itch caused by life-saving medications.

At the heart of their discovery is a type of sensory nerve cell whose endings receive information from the skin and relay it to other nerves in the spinal cord, which then coordinates a response to the stimulus. Published online Dec. 23 in Nature Neuroscience, a report on the research suggests that even when the itch-specific nerve cells receive stimuli that are normally pain-inducing, the message they send isn?t ?That hurts!? but rather ?That itches!?

Pain and itch are both important sensations that help organisms survive. And pain is arguably more important because it tells us to withdraw the pained body part in order to prevent tissue damage. But itch also warns us of the presence of irritants, as in an allergic reaction. However, ?when either of these sensations continues for weeks or months, they are no longer helpful. We even see patients stop taking life-saving medications because they cause such horrible itchiness all over,? says Xinzhong Dong, a Howard Hughes early career scientist and associate professor of neuroscience at the Institute of Basic Biomedical Sciences at the Johns Hopkins University School of Medicine. ?And sometimes when we try to suppress chronic pain, with morphine for example, we end up causing chronic itchiness. So the two sensations are somehow related, and this study has begun to untangle them,? he says.

Because nerve cells send their messages as electrical currents that flow through them just as they would through wires, scientists can plug tiny monitors into individual nerve cells to detect the moment of stimulation. The scientific controversy over pain and itch centers around a group of nerve cells known to respond electrically to painful stimuli such as molecules of capsaicin, the fiery ingredient in chili peppers. A small subset of these nerve cells also responds electrically to itchy stimuli because they have on their surfaces receptors for molecules like histamine. One of these itchy receptors, called MrgA3, binds the anti-malaria drug chloroquine, causing serious itchiness in many patients.

Sensory nerve scientists have not known whether the nerves with itchy receptors and pain receptors were actually sending both types of messages to the brain, or just itch messages. What the current study found is that, in nerves with the itchy receptor MrgA3, electrical signals sent in response to both painful and itchy stimuli are interpreted by the brain as itch.

To reach this conclusion, the researchers first used a genetic trick to label the MrgA3 cells in mice with a glowing protein that allowed them to see the cells under the microscope. Aided by the glow, they were able to plug in those tiny electricity monitors and watch nerve cell responses to different stimuli. The cells transmitted electrical signals when the mice were exposed to itch-inducing chloroquine and histamine, as well as pain-inducing capsaicin and heat. Based on this result, the researchers tentatively concluded that the cells could send both pain and itch signals.

In the next experiment, the researchers monitored the behavioral responses of mice to the different stimuli. As expected, when the tails of normal mice were placed in hot water, they quickly pulled them out; when normal mice were given a bit of chloroquine or histamine, they scratched vigorously with their hind legs.

Then, to examine the role of MrgA3 cells in pain and itch, the scientists selectively killed MrgA3 nerve cells in adult mice and retested their responses. Presumably, the researchers noted, because MrgA3 cells are only a small fraction of all pain-sensing nerve cells, the mice had normal withdrawal responses to painful stimuli like hot water. However, when exposed to itchy stimuli, their scratching responses were reduced to varying degrees depending on the stimulus, most significantly in response to chloroquine. The fact that some stimuli still caused scratching suggested to the scientists that MrgA3 cells are not the only ones in the body that respond to itch. ?We were convinced that MrgA3 cells are responsible for much of the sensation of itch, but it wasn?t yet clear whether MrgA3 cells could also relay painful information,? says Dong.

In their final experiments, the scientists used genetic techniques to create mice in which the MrgA3 cells were the only cells in the body capable of responding to capsaicin, that peppery pain-inducing substance. When injected into the cheeks of mice, normal mice massage the area with their forepaws to relieve the hot sensation. When injected into the experimental mice, they vigorously scratched their cheeks with their hind legs, suggesting that this normally painful stimulus had been communicated to the brain?by MrgA3 cells?as itchiness.

"Now that we have disentangled these itchy sensations from painful ones, we should be able to design drugs that target itch-specific nerve cells to combat chronic itchiness," says Dong. "We hope that this will not only provide relief, but also increase people's faithfulness to their drug plans, particularly for deadly diseases like malaria and cancer."

Determined to make it 3-0 says Ed Cowan ? Cricket News Update

Australian Test opener, Ed Cowan, says that the team is motivated to give a fitting send off to Michael Hussey by securing a whitewash against Sri Lanka when both sides meet in Sydney.

The team?s preparations have been hindered by various issues, which also showed in their training which Cowan himself felt to be ?pretty flat?. Though the dilemma surrounding the captaincy issue cooled down after skipper Michael Clarke took part in the full training, which makes him certain for the upcoming clash, the larger picture seems bleak for the national team.

Just as the team was recovering from the loss of Ricky Ponting, their all-time leading run-scorer, news floated that Hussey has also made up his mind to bid adieu to international cricket as he prepares to spend more time with his family. That leaves a gaping hole in Australia?s middle-order, and changes some ground rules for the selectors?, who are now aiming to add a wiser head to the team?s roster rather than putting faith in a youngster.

While the upcoming match holds little importance, Cowan was keen to outline his side?s ambitions where they will look to clean sweep their opponents, which will help them erase the disappointing result earlier this month where they lost to South Africa.

"We have spoken at some length about our determination to make sure we win this series 3-0," Cowan said. "We experienced some disappointment earlier in the season, and this is us trying to vent that a little bit and we are pretty keen to see Mike off with flying colours."

The opener also downplayed suggestions that the SCG track will aid the side in preparing for their upcoming tour of India.

"It's not like we're flying out [to India] tomorrow and are dumped in the middle of Hyderabad and told good luck. It's six weeks [away]. I'm pretty keen to face spin [in the Sydney Test] if the wicket looks like that, because I don't think it's going to turn very much, so if Sri Lanka want to play two spinners that's probably to our favour," the Tasmanian added.?

Gambian leader says to build herbal AIDS-cure hospital

BANJUL (Reuters) - AIDS patients would be offered an herbal cure at a 1,111-bed hospital in Gambia that the president said on Tuesday he plans to build despite medical concerns the treatment is dangerous.

President Yahya Jammeh said in 2007 he had found a remedy of boiled herbs to cure AIDS, stirring anger among Western medical experts who claimed he was giving false hope to the sick.

"With this project coming to fruition, we intend to treat 10,000 HIV/AIDS patients every six months through natural medicine," Jammeh said in his New Year's address, adding that he expected the 1,111-bed hospital to open in 2015.

The World Health Organisation and the United Nations have said Jammeh's HIV/AIDS treatment is alarming mainly because patients are required to cease their anti-retroviral drugs, making them more prone to infection.

Jammeh said in October that 68 HIV/AIDS patients undergoing his herbal remedy had been cured and discharged, the seventh batch since the treatments began five years ago.

Other African leaders have drawn criticism for extolling the power of natural remedies to combat AIDS.

The administration of former South African President Thabo Mbeki was ridiculed for denying there was a link between HIV and AIDS while prescribing meaningless treatments such as beet root instead of internationally proven medicines.

The HIV rate in Gambia is relatively low compared to other African states, with 2 percent of the country's roughly 1.8 million people infected, according to the United Nations.

Jammeh came to power in Gambia, a sliver of land on Africa's west coast that is popular with sun-seeking European tourists, in a bloodless military coup in 1994.

He is accused by activists of human rights abuses during his rule, and most recently drew international criticism for executing nine death-row inmates by firing squad.

Dark chocolate craft beer cake with dark chocolate frosting | Food ...

An easy way to make sure that your cake is tender and not too dense - whip the egg whites separately to the egg yolks and fold them in at the end. While the extra step is a bit of a pain, the end results are TOTALLY WORTH IT (and worthy of the capitalized letters!).?

Being a shareholder in a start-up beer distributor means that we have a lot of great craft beer around. While I love to drink beer, I can only drink a glass before getting full. My preference is to cook with it. I find that using beer in cooking imparts a certain fullness of flavor to the food but is still subtle enough not to overpower it. I've made Irish beef stew with Tokyo black porter, salted caramel ice cream with Rogue Hazelnut Nectar brown ale. However, one of the recent standouts has been this decadent, rich dark chocolate cake made with Tokyo black porter, but would likely also work well with any other stout, porter or good dark beer. I plan to make it with Rogue Mocha Porter soon.?

This chocolate cake is extremely rich and dense, with a thick chocolate ganache frosting. A small slice would go very well with coffee (or ice cream). Top with your favorite fruit or candy - or leave plain. While you can splurge and use the best dark chocolate to make this cake, I find that the 100g bars of chocolate such as Lindt works fine. (I like to use the cheaper dark chocolate sold in Ikea food mart - it's half the price of Lindt and has a very good texture and taste. I've also used Green & Black chocolate too for special occasions.) I suppose you could also use baking/unsweetened chocolate too, though I can't guarantee the results. Chocolate is the star of this cake, so you want to use the top quality chocolate you're willing to pay for.?

The full amount for this recipe below makes one round 9-inch layer cake (using two 9-inch round pans), or you can halve the ingredients to make a 9-inch square cake.?

Bon App?tit!

Ingredients

Cake:?

  • 3 oz (85g) dark chocolate (preferably min. 60% cacao)
  • 2 cups all-purpose flour?+ 1/4 cup unsweetened cocoa powder (200 g flour?+ 50g cocoa powder)*
  • 2 tsp baking powder
  • 1/2 tsp salt
  • 200g (1 3/4 sticks) butter (room temperature)
  • 200g (1 cup)?+ 3 tablespoons caster/granulated white sugar
  • 3 large eggs, separated
  • 3/4 cup stout, porter or dark beer such as Rogue Mocha porter or Tokyo Black porter
  • 1/2 cup espresso or strong coffee (cooled)

* Cocoa powder is optional. If you don't have it, just make up the difference with the flour.?

?Frosting:?
  • 450g (1 lb) dark chocolate (50-60% cacao)
  • 2 cups heavy whipping cream (I used UHT whipping cream because it's hard to find fresh cream in Hong Kong)

Method:

Cake:??

  1. Preheat oven to 350F or 180C.?
  2. Butter and use baking paper line your two round 9-inch cake pans (or one pan if using a half-recipe). Set aside.?
  3. Melt the chocolate for the cake in a bain-marie?(put a metal bowl on top of a saucepan of barely simmering water), or microwave it. See here for a quick guide to melting chocolate. Set aside.?
  4. Whisk together the dry ingredients (flour, cocoa powder, baking powder, salt). Set aside.?
  5. Using a mixer, beat the egg whites and 3 tablespoons sugar until stiff but not dry. In layman's terms, this means that the egg whites look like meringue and stay in the bowl (they don't slosh about or fall when you tip the bowl sideways or upside down). Here's a good Chowhound thread about beating egg whites. Set aside.
  6. Wipe clean your beaters and use another bowl for the butter and sugar. Using a mixer, cream (beat together) the butter and sugar until fluffy and pale yellow. Add egg yolks 1 at a time, beating well after each addition. Beat in melted chocolate, then the beer and coffee.?Add in flour in 2 additions until incorporated. (If you overmix the batter, your cake crumb may be coarse and tough.)
  7. Gently fold in your egg whites in 2 to 3 additions to the batter. Don't overmix the batter to avoid dissolving the egg whites - the air in the beaten egg whites helps to lighten the texture of the cake!
  8. Scrape the batter into pan, then bake in oven until cake tester comes out dry (about 30 minutes).?
  9. Remove from oven and transfer to cooling racks.?
The cake can be made ahead 1 day before frosting and assembly. (Cover and store and room temperature. Or, if you're really busy, make it ahead up to 1 week in advance and freeze the cake layers individually. Defrost it before adding frosting.)

Frosting:?

  1. Placed chopped chocolate in a metal bowl.???
  2. Heat cream in a saucepan until simmering.?
  3. Pour hot cream over the chocolate.?
  4. Wait one minute, then whisk together until mixture is smooth.
  5. Chill in fridge until thickened and spreadable (2 hours in fridge or 30 minutes in freezer).?

The frosting can also be made ahead separately and stored in the fridge up to 3 days before assembling your cake.?

Assembly:

  1. Frost your cake, then top with your favorite toppings (white or other colorful toppings are a good contrast for the dark chocolate). For this winter birthday cake, I used white chocolate bark, white chocolate Maltesers, fresh pomegranate seeds, and diced fresh red pear.
  2. For layer cake, you may need to use a serrated knife to level off the top of the cake in order to get flat even layers.?
  3. For single-layer sheet cakes, you can stick the entire cake (wrapped/covered in plastic wrap) in the freezer for up to two weeks. I highly doubt it would last that long, though!

GigaOM: Intel Media TV service won't launch at CES, but it is coming soon

GigaOM Intel Media TV service won't launch at CES, but it is coming soon

We've seen several reports over the last week about Intel's set-top box efforts, and an impending CES launch. According to GigaOM, some of those rumors simply aren't true. The site has confirmed with several sources at Intel that a set-top box (and a whole lot more) is in the works, but a CES announcement has not been planned. Instead, Media, the mysterious Intel division, which is run like a startup with staffers hired from other Silicon Valley companies rather than shifted from different departments, could be unveiled in March, or perhaps even at the AllThingsD Dive Into Media conference in February.

A detailed GigaOM report outlines the company's plans, which do include a STB manufactured by Intel that would compete with Apple TV, but also solutions for other platforms, including computers, tablets and smartphones. According to GigaOM sources, the company has spent more than $100 million on Intel Media, so it's clearly taking the effort seriously. Still, until Intel Media has been exposed to the market, it's too soon to judge its success. Hit up our source link for the full scoop.

GOP leader Cantor opposes Senate 'cliff' bill

WASHINGTON (AP) ? The No. 2 Republican in the House leadership says he opposes a Senate-passed measure to avert the so-called fiscal cliff.

Virginia Rep. Eric Cantor told reporters after a two-hour closed-door meeting Tuesday with his GOP lawmakers that he did not support the bill.

He said House leaders were looking for "the best path forward" and that no decisions had been made.

The Senate passed the measure early Tuesday by a sweeping 89-8 vote. House passage of the measure would send it to President Barack Obama for his signature. The bill would increase taxes on family income exceeding $450,000 and delay across-the-board spending cuts for two months.

House GOP leaders were considering amending the measure and sending it back to the Senate, but that step could produce a deadlock.

Broncos get top seed in AFC

Thanks to Houston's late-season slump, Denver and New England will have byes when the AFC playoffs begin next week.

The Texans fell from first to third in the conference Sunday when they lost 28-16 at Indianapolis, which welcomed back coach Chuck Pagano after nearly three months of treatments for leukemia.

AFC West champion Denver won its 11th straight game, 38-3 over Kansas City to secure the top seed. New England blanked Miami 28-0 for the second spot.

Minnesota edged Green Bay 37-34 to grab the final NFC wild card, sinking the Packers to the third seed. Those teams will meet again next Saturday night at Lambeau Field.

The other NFC matchup will have Seattle (11-5), which beat St. Louis 20-13, at either Washington or Dallas on Sunday at 4:30 p.m. ET. Those teams met Sunday night for the NFC East crown.

Cincinnati (10-6) will be at Houston on Saturday at 4:30 p.m. ET, and Indianapolis (11-5) goes to at Baltimore (10-6) on Sunday at 1 p.m. in the AFC wild-card rounds.

The divisional round games will be hosted by Denver on Saturday, Jan. 12, followed by San Francisco at night. On Sunday, Jan. 13, Atlanta will host the early game, followed by New England.

Peyton Manning threw for three touchdowns as Denver (13-3) routed the Chiefs. New England (12-4) got the second seed despite having the same record as Houston because it beat the Texans, who lost three of their final four games.

Adrian Peterson had 199 yards against the Packers, finishing with 2,097 ? Dickerson's single-season rushing mark in 2,105. But it was rookie kicker Blair Walsh who won it with a 29-yard field goal as time expired.

"''Ultimately we got the 'W,'" Peterson said.

Baltimore Pro Bowl safety Ed Reed is looking forward to a reunion with Pagano. He wishes it would come a little later in the postseason.

"Chuck's like a dad to me," Reed said "He means a lot to me. I would have much rather seen them in the AFC championship game than the first game."

But Reed will see him next week at Baltimore.

The Ravens had a chance to move up to the AFC's third seed with a win and a New England loss. But Baltimore lost at Cincinnati as both teams played backups for much of the game.

Pagano coached the Ravens' secondary for three seasons and was promoted to coordinator last year. Players and coaches in Baltimore have kept in touch, offering encouragement as he fought through the cancer treatments.

"Going back to Baltimore, obviously there's some familiarity there," Pagano said. "We had four great years there as a family. It's a top-notch organization, you know, really good football club. It's a great challenge and they have a great team and they have great players all over the place."

The Colts were 2-14 last season and chose quarterback Andrew Luck with the top selection in the draft. Luck and offensive coordinator Bruce Arians, who stepped in as interim coach with Pagano sidelined, led the turnaround.

Next week, Pagano goes up against former boss John Harbaugh.

"I love his family, and he's one of my closest personal friends in coaching," Harbaugh said. "What he's been through is phenomenal, but we're all competitors so that gets set aside."

The defending Super Bowl champion Giants are out of contention. When Chicago beat Detroit 26-24, the Giants (9-7) were eliminated, even though they routed Philadelphia 42-7.

"It hurts," said Eli Manning. "Each year you want to make the playoffs to give yourself an opportunity to win a championship; 9-7 last year was good enough. It wasn't good enough this year and we knew it wouldn't be."

Minnesota's win eliminated Chicago, which the Vikings swept this season.
